Separation of aldobiuronic acids by an anion exchanger

Abstract: The sepn. of aldobiuronic acids from the uronic acids on strongly basic anion exchanger Dowex 1-X8, 100-200 mesh, in the acetate form, and with a gradient HOAc elution, was described. The aldobiuronic acids were eluted first and then the uronic acids. 6-O-(4-O-methyl-β-D-glucuronopyranosyl)-D-galactose, and 6-O-(β-D-glucuronopyranosyl)-D-galactose were detd. by elution with 0.2M HOAc. 2-O-(β-D-Glucuronopyranosyl)mannose, a mixt. of 4-O-methyl-D-glucuronic acid and D-glucuronic acid, 4-O-(α-D-galacturonopyranosyl)-D-galactose, D-galacturonic acid, and 4-O-(α-D-galacturonopyranosyl)-D-galacturonic acid were detd. by elution with 0.4M HOAc. Further sepn. of the mixt. could be carried out by paper chromatography. These methods of preparative sepn. were applied in structural investigation of the gum in Prunus armeniaca and pectic acids in Malus sylvestris.
